  Yes you can. When you unlock your Playthrough 2 everything is reset and made harder so you can get a chnace to do everything again.

  Yes you can but beware your level # when do this. You begin the game in 0 level and the zombies are 10. So if you finish the game (1st play through) in 34, you have to rank your character up to 44 in order to play it again without problems.

  actually dlc is the only fast travel areas you cant find and the only ones available even at level one before you fix the travel thing.... i must beat your ps3 over the skull for this. :D

  you have to progress in the game in order to get there, so just keep playing till you find it
